Podcaster and commentator Joe Rogan has often spoken publicly about his extensive supplement regimen. He’s particularly fond of anti aging supplements and has stated that he takes nicotinamide mononucleotide (NMN) and resveratrol to enhance longevity, energy, and mental focus. While these are both highly prized anti aging supplements, they are far from the only ones on the market.

NMN is a naturally occurring molecule in the human body that serves as a precursor to the coenzyme nicotinamide adenine dinucleotide (NAD+). NAD+ plays a role in many vital biological processes, including metabolism, cell growth, and DNA reproduction. As people age, NAD+ production naturally declines, which can make it more difficult for the body to undertake these cellular processes effectively. NMN supplements purport to address this issue, giving the body the necessary tools required to synthesize more NAD+ for increased energy production. NMN has been shown to increase blood plasma NAD+ levels; however, this is not always mirrored in muscle tissue. Moreover, NMN was recently banned by the FDA in the USA.

Resveratrol has shown some therapeutic effects, such as improving some oxidative stress markers; however, it has mixed results in more functional outcomes, such as endothelial (vascular) function, and human trials are still limited, so more research is needed.

In contrast, MitoQ has undergone extensive research to confirm its effectiveness and methods of action. Mitoquinol mesylate is a modified form of coenzyme Q10 (CoQ10). MitoQ has been extensively studied for its unique ability to penetrate the mitochondrial membrane, providing direct support to mitochondria. Its antioxidant-improving effects have been shown clinically via biomarkers such as catalase, and it also has been demonstrated to support vascular function by increasing arterial flexibility.

Bioavailability

Even the most effective, expensive supplements are effectively wasted if they’re not properly absorbed by the body. This is referred to as bioavailability, which measures how much of an ingested substance is able to enter the bloodstream and distribute throughout the body. NMN, resveratrol, and MitoQ have all been subject to studies on their bioavailability.

NMN has been found to have poor absorption, and most resveratrol is transformed into other molecules once it’s absorbed into the digestive tract, so the original form of resveratrol is found in very low levels in the blood. MitoQ, however, has exceptional absorptive potential and also stays in the same form throughout the body. A study has found that it can penetrate the mitochondrial membrane, directly supporting the powerhouse of the cell where it’s most needed.

Benefits for the circulatory system

Both resveratrol and MitoQ claim to benefit heart health. Resveratrol has been found to have the potential to prevent atherosclerosis or the hardening of the arteries; however, this was only shown in a diabetic population – so its performance in healthy populations remains uncertain. MitoQ has undergone many in vivo trials – performed with healthy people – and one study found that MitoQ may support circulatory health by improving vascular function. Resveratrol’s results are conflicting when it comes to heart health, with some studies showing a slight effect while others showed no effect. In contrast, MitoQ has been studied extensively in vivo and has demonstrated supportive heart health functions in several studies with normal (non-diseased) subjects.

Benefits for metabolism

Animal studies have suggested that resveratrol can improve metabolism but have not found significant evidence of resveratrol’s potential to reduce body weight or change body fat distribution in healthy humans. Overall, the results regarding resveratrol’s effects on metabolism are varied and inconclusive. MitoQ has been shown to increase expression of PGC1-alpha, a master switch of metabolism, but more studies need to be done to understand MitoQ’s overall effects on weight or body composition.

Retaining strength and mobility as you age is a key indicator of longevity. While some studies have shown that resveratrol supplements can reduce muscle damage from exercise, there is some evidence that it could blunt the positive effects of exercise training on cardiovascular health in aged men. NMN improved some aspects of aerobic performance, such as Vo2MAX, in amateur runners, but only at higher doses (e.g., 600-1200 mg).

One MitoQ study has shown that six weeks of supplementation (20mg) in middle-aged and older adults support mobility with age. The 2018 trial by the University of Colorado found that MitoQ increased leg extension power, a key indicator of mobility and strength as you age. Other trials have shown MitoQ aids exercise performance, helping with muscle power and output and the ability to safeguard against DNA damage after intense exercise.
